چکیده

Techniques for sputum homogenization and quantitative cultural methods have enabled clinical microbiologists to make a more objective assessment of the bacterial flora found in lower respiratory tract infection. This paper describes the application of the Colworth Stomacher for rapid and effective homogenization of sputum and the use of a calibrated loop for quantitative culture of the homogenate. The Stomacher (A. J. Seward, London SEI 9U6) has been designed for the blending of powders, liquids, samples for microbiological analysis, emulsification, and extraction. A suitable plastic bag containing the sample is placed in the stomaching area and sealed near the top by the closed Stomacher door. Two paddles alternately squeeze the bag and its contents against the inner aspect of the door (Sharpe and Jackson, 1972).

Development and Evaluation of Real-Time Reverse Transcription Polymerase Chain Reaction Test for Quantitative and Qualitative Recognition of H5 Subtype of Avian Influenza Viruses

Avian influenza viruses (AIV) affect a wide range of birds and mammals, cause severe economic damage to the poultry industry, and pose a serious threat to humans. Highly pathogenic avian influenza viruses (HPAI) H5N1 were first identified in Southeast Asia in 1996 and spread to four continents over the following years. The viruses have caused high mortality in chickens and various bird species ...

Quantitative sputum culture as a means of excluding false positive reports in the routine microbiology laboratory.

A relatively simple technique for sputum homogenization and dilution is described. Results show that this technique is reliable for isolation and quantitative culture of organisms from cases of chest infection. The technique has resulted in significant reduction in false positive reports, particularly when a system of interpretative reporting is utilized based on 10(7) organisms per millilitre ...

Quantitative Bacterial Micro-Assay for Rapid Diagnosis of Galactosemia: Application in Galactosemia Neonatal Screening

In the present study a new economic and rapid bacterial micro-assay for simultaneous detection and quantitative measurement of serum galactose was developed. Analysis of the standard curve showed a linearity range for galactose from 2 mg/dL to 180 mg/dL with a regression equation of Y = 0.013X ? 0.083; R² = 0.962. The advantage of the method is its ability to measure serum galactose quantitativ...

Rapid quantitative detection of Listeria monocytogenes in chicken using direct and combined enrichment/qPCR method

Listeria monocytogenes is a species of foodborne pathogen often related to foods, such as poultry, ready-to-eat products, fruits, and vegetables. The culture method is a standard procedure for the detection of bacteria in food products. The real-time quantitative PCR (qPCR) technique can be used for the quantification of foodborne pathogens. The current research was aimed to assess...
